Development of rapid diagnostic kits for aquatic disease-acute hepatopancreatic necrosis(EMS) detection strips/FRI

Early mortality syndrome (EMS), or acute heptaopancreatic necrosis(AHPND) was officially reported in China. The disease signs associated with mortality typically appear within 20 to 30d after stocking ponds with postlarvae. The syndrome of this disease is lethargic sleep and anorexia. Researchers can obverse that the weakness shrimp would assemblage on the bottom of culture pond, and they usually have soft shell and wriling swimming. Shrimp suffering from EMS/AHPND show significant atrophy in hepatopancresas, pale to white hepatopancresas and no gut contents. In Taiwan, so far we have not find any reporters for shrimp infocted EMS or acute heptaopancreatic necrosis. But should prevent it happening.
A rapid diagnosis kit that dectects AHPND antigens with an immunochromatographic strip (ICS) test was developed in this study, and it will be used for aquaculture farm. The ICS test was produced using colloidal gold coated with polyclonal antibodies (pAbs) against Vibrio parahaemolyticus. Two pAbs were developed from rabbits immunized with extracted protein of Vibrio parahaemolyticus. Based on the two pAbs pairs, two types of ICS were prepared. The strips is named Vp6strip. V6strip showed a higher diagnostic sensitivity and specificity and the minimum limitation of ICS sensitivity indicated 1.0x10^5 colony forming units (CFU) and antigen concentration of Vibrio parahaemolyticus is 0.1ug. In the sensitivity test, Vp6strip can detect four pathogenicity Vibrio parahaemolyticus which caused AHPND and the assay time for dection of Vibrio parahaemolyticus antigens is less than 15 min.
